Immunologic protection of rabbit corneal allografts: preparation and in vitro testing of heterologous "blocking" antibody.

چکیده
Corneal allografts, like other foreign tissues, are subject to immunologic attack and rejection. Efforts to prolong graft survival have included the use of antilymphocyte serum and blocking antibodies. In this study antilymphocyte serum was tested as a potential corneal allograftblocking reagent. It has been demonstrated that guinea pig anti-rabbit lymphocyte serum and the gamma globulin fraction of this serum bind to rabbit lymphocytes and corneal cells. In the presence of serum complement the antiserum and globidin are cytotoxic for cells of the cornea. Chemical modificaton of the anti-lymphocyte globulin renders it incapable of complement fixation, creating a reagent which retains the capacity to bind to cells and, in addition, to protect them from the cytotoxic effects of unaltered antibody and serum complement. The benign and protective nature of modified antilymphocyte globulin suggests that it is an excellent candidate for use in the prevention of corneal graft rejection.

Heterologous Expression of Potato Virus Y Coat Protein, Isolate Pot187
Background: The advent of recombinant DNA technology has facilitated heterologous expression of proteins from various sources in different host systems including Escherichia coli. If a plant virus coat protein is expressed in the bacterium it can be used as the antigen for antibody preparation. Such a recombinant antigen preparation can be particularly useful where equipment such as ultracentri...
متن کاملPromotion of murine orthotopic corneal allograft survival by systemic administration of anti-CD4 monoclonal antibody.
A mouse model of orthotopic corneal allograft rejection was used to examine the efficacy of anti-CD4 and anti-CD8 monoclonal antibodies in preventing immunologic rejection of corneal allografts. Although it is believed by many that corneal graft rejection is mediated, at least in part, by CD8-positive cytotoxic T-lymphocytes, systemic administration of anti-CD8 antibody did not reduce the rejec...
متن کاملCell-mediated immunity Immunologic privilege Rabbit Mouse Rat Immunosuppression
The major findings regarding corneal allograft rejection in experimental animals are reviewed. The principal anatomic and biological feature of the cornea that determines the immunologic privilege of this tissue is its avascularity. The surgical trauma of transplantation compromises the immunologic privilege, putting corneal allografts at risk for immune rejection. During the past 50 yr, rabbit...
متن کاملSome aspects of the immunologic factors in corneal grafts.
The general basic problems of homotransplantation are briefly reviewed; and pertinent aspects of the factors involved are applied to the corneal graft problem. It is pointed, out that one of the most important questions yet to be solved is the isolation and characterization of the antigens involved. The necessity for an in vitro technique to study this problem prompted the preparation of antico...
متن کاملGalectin-9-Mediated Protection from Allo-Specific T Cells as a Mechanism of Immune Privilege of Corneal Allografts
The eye is an immune-privileged organ, and corneal transplantation is therefore one of the most successful organ transplantation. The immunosuppressive intraocular microenvironment is known as one of the mechanisms underlying immune privilege in the eye. T-cell immunoglobulin and mucin domain (Tim)-3 is a regulatory molecule for T-cell function, and galectin (Gal)-9 is a Tim-3 ligand. We invest...
